High youth unemployment and cutbacks in government funding for post-secondary education are the new realities confronting students. The implications for most working and middle class students are either to abandon education altogether and to accept a future of McJobs and unemployment, or to be saddled with a lifetime of debt. Since the end of WW II, science and technology have been playing a dramatically increased role in the process of capitalist production in Canada. For example, between 1971 and 1986, jobs primarily concerned with the creation and utilization of data and technical knowledge have represented two-thirds of net job growth. This has led to a rapid expansion in school enrolment. Between 1951 and 1993, the number of full-time post-secondary students has increased over tenfold, growing from 91,000 to almost 1 million. However, accompanying the scientific and technological revolution, is a tremendous growth in productivity that has led to higher levels of unemployment. Between 1980 and 1993, youth unemployment increased from 12% to 17.5, leaving many with no other alternative but to remain in or return to school. Since 1984-85, tuition fees have more than doubled across Canada, pushing more students into the labour market in search of income. While in 1980, 31% of full-time students, aged 15-24, held jobs during the school year; by 1989, this number had reached 41%. This worsening economic situation has also seriously interfered with the studies of college and university full-time students, as almost a third of them were forced to work 20 or more hours per week during the school year. Another reflection of the economic hardship of students is their growing debt burden. In 1984, 114,000 Ontario students received around $4000 million in student aid, by 1993-94, as tuition continued to increase, about 180,000 students (representing almost half of full-time students) took out over $1 billion in loans. The average value of loans in 1994-95 was around $6800. By 1998, the federal government is scheduled to eliminate a total of $7 billion in transfer payments to the provinces for medicare, social assistance, and post-secondary education. The expected effect on Ontario university students is a doubling of their tuition. But this is only the tip of the iceberg. Ontario’s education minister, John Snobelen, has already made clear his government’s intention to move toward a â€Å"market-based tuition fee†. If both levels of government abandon their financial support for Ontario universities, tuition will reach minimum levels of $7500-$8000. To counter the danger of a dramatic growth in defaults as it increases tuition fees, the federal government has transferred liability for student loans to the private banks. While increasing the role of the private banks in the short term, these changes set the pace for the full privatization of the student loan system. Another plan, currently under discussion, proposes collecting the loan repayments through the taxation system, i.e., Revenue Canada, through an Income Contingent Loan Repayment Plan (ICLRP). If the direction of government â€Å"reforms† is not reversed, it is only a matter of time before students who are at a high risk of unemployment (disproportionately women) or who are expected to have a low income after graduation (again disproportionately women) are refused access to loans, while most of the rest are burdened with a lifetime of debt. But the class nature of the privatization process has already become apparent. For 1995-96, the demand for first-year places is down by 5% in Ontario–20% in some faculties–leading many schools to turn toward recruiting drives and to lowering their admission requirements. Evidently, higher tuition is an obstacle for better qualified, poorer students, while offering an opening to less qualified, richer students. Cut the deficit by taxing the big corporations By focusing attention on expenditure reduction, the big corporations and the right-wing aim to deflect attention from the cause of the national debt: the decreasing tax revenues from corporations, which have declined from 20% of total federal revenues to only 7% in the last 20 years. Canada continues to have one of the lowest corporate tax rates in the industrialized world. Even the same level of corporate taxation as in the U.S. would provide an additional $9 billion a year–more than enough to cover all the cuts in social spending scheduled by the federal government. The ongoing cutbacks to education follow more than a decade of political and financial pressures promoted by federal and provincial governments in relation to social spending in general. Indeed, the education crisis cannot be separated from the overall crisis of the capitalist economy in Canada. The critical challenge before us today is to bring together all the social elements that are hurt by the implementation of the big corporate agenda: to build wide coalitions of youth and students, workers, women, seniors, environmentalists, peace groups, farmers, aboriginal people, immigrants, and many others–in support of a genuine â€Å"People’s Alternative† program. This program would provide for decent wages, stronger public health and child cares systems, job creation programs, while restoring and increasing public funding for education. Reversing the current cut-backs and building the people’s alternative depend on escalating the student protests. As a first step, we must build strong grassroots organizations on campuses which can ensure wide student involvement in the mobilizations. Students and working people have fought a long battle to win the right to higher education, health care, UI, and other social programs. The Next New Thing PersonalizedLearning One thing about education as a profession- the grass is never allowed to grow under a teacher’s feet for very long. There’s always something new coming down the pike, which isn’t necessarily a bad thing, but it does induce a kind of professional dizziness from time to time. Adapting to the â€Å"next new thing† is always a challenge, especially if it moves from the periphery onto the center stage in classroom practices. The most recent concept in education is personalized learning, not to be confused with differentiated instruction. Differentiated instruction addresses how lessons are delivered to students, based on their individual needs. Personalized learning is more complex. In its purest form, personalized learning allows students to choose what they will study and to proceed at their own pace in meeting individual goals. Implementing it requires four instructional strategies, which are summarized  in â€Å"Personalized Learning: A Working Definition†: Continual assessment as a student progresses toward â€Å"clearly-defined goals,† with the student advancing and earning credit after demonstrating mastery. Learning environments designed to meet students’ needs in reaching their goals. Customized  Ã¢â‚¬Å"learning paths† based on each student’s progress, motivations, and goals. Up-to-date â€Å"learner profiles† that chart each student’s strengths, needs, motivations, and goals. That’s a lot to do after taking attendance! Seriously, it’s a lot to do period, and the current demands of standardized  testing make personalized learning seem like a classroom fantasy. It’s a great idea, though, so what can be done to personalize learning in a system based on covering a specific curriculum in a limited amount of time?  Apparently, quite a bit, starting with learner profiles. The learner profile is one part of personalized learning that’s already in place. Learner profiles may not be written down, but they exist,  filed  in teachers’ minds where they’re continually reviewed and updated. Ask a teacher about particular students, and be prepared for  a  detailed  report on their individual strengths, weaknesses, and motivations. (You’ll also probably find out what’s holding them back or pushing them ahead and who frequently needs to borrow lunch money.)  I think we’re good on learner profiles, except perhaps for having students set some specific learning goals  for themselves. The rest of personalized learning has been broken down into five essential elements: Choice, Pace, Path, Mode, and Data.  Within these five elements, there’s room to personalize learning even when a teacher is tied to an articulated curriculum and a pacing guide in meeting state standards. Choice and Pace Be creative in writing curriculum-based lesson plans so that they offer students some choice in what they will study. Let students choose their own topics for research papers, allowing them to study what most interests them. Create a list of activities that address students’ interests, different learning styles, and multiple intelligences, and allow them to choose their own activities from the list. Design an independent study project with several parts, and let students decide how they will accomplish each part- what they will read and write and which project activities they will complete. Let students design and pursue their own independent study projects. Considering the time constraints in a classroom, it’s unlikely that all students will be able to proceed at their own pace, all the time, in completing their work, but it’s usually possible to be somewhat flexible in setting due dates for assignments. Path and Mode These elements refer to the ways individual students pursue learning objectives and the means they use in mastering them. Providing students with different learning paths and different modes of study requires making some fundamental changes in instruction. Everyone won’t be doing the same thing at the same time. That sounds like a recipe for chaos, but not necessarily! Check out these examples of how to reorganize instruction efficiently and effectively in personalizing learning, and see  four more K-12 examples here.  Technology tools and the resources that can be tapped through them can play an essential role in personalized learning. Edutopia offers ten tips for using technology in implementing it. Data Regardless of how learning takes place, it has to be assessed. The prospect of writing individual tests for every student is daunting indeed, and actually doing it is most likely impossible. However, there are more ways than testing to assess learning. Learner Centered Teaching  lists dozens of ways to do it without giving tests, and using only tests in assessment is a bad idea anyway. One-size-fits-all never works well, so it’s not surprising that personalizing learning is the ideal way to educate kids. The challenge is figuring out how to accomplish it while simultaneously meeting the demands of †¦ well, everybody. It’s easy to think, â€Å"Personalized learning- can’t be done!† It’s true that personalizing every student’s learning, all the time in every way, is probably not possible in today’s classrooms, but there’s plenty of wiggle room in the concept. Exploring how to make the most of it so that kids achieve more and learn in ways that are more natural and meaningful for them is a good use of planning time. Functional Multi-enzyme Complexes In Vitro Molecular self-assembly offers a method of sophisticated materials constructed with precision. Designing self-assembling enzyme structures is of particular interest for the unique functional capabilities of enzymes, as shown in Figure 2. Chemically induced assembly has been shown to be a powerful tool for the investigation of cellular events and for its easy operation and low cost compared to bioconjuction. Chemical inducers can be cofactors, inhibitors, metal ions, which are based on specific interaction of molecule and enzyme. Chemical inducers bring the two enzymes together to form multi-enzyme. A number of reviews have covered the self-assembly of proteins and enzymes by chemicals. King N.P. et al discussed the principles employed in recent efforts to design complex and geometrically specific protein assemblies, with a focus on practical approaches. However, precise manipulation of protein self-assembly behavior in vitro is still a great challenge. Here we review recent studies i n the chemical induced self-assembly of multi-enzyme system from the perspective of multi-enzyme complex organization, enzyme interactions, and regulation of assembly. Inhibitor induced multi-enzyme assembly Inhibitor induced dimerization has been reported as the controlled dimerization of proteins via dimerizers. During the process of dimerization, the dimerizers assemble proteins into homospecific or heterospecific multivalent nanostructures. An enzyme inhibitor binds with enzymes specifically and decreases their activity. Drug discovery typically focus on the identification and design of inhibitors to perturb enzyme function, which greatly depend on the chemical structure. Carlson and co-workers reported self-assembly of wild-type Escherichia coli dihydrofolate reductase (DHFR) into protein nanorings using dimeric methotrexate molecules, which tethered together by a flexible peptide linker. The enzymes are capable of spontaneously forming highly stable cyclic structures with diameters ranging from 8 to 20 nm. The nanoring size is dependent on the length and composition of the peptide linker, on the affinity and conformational state of the dimerizer, and on induced protein-protein interactions. Chou reported the preparation of dihydrofolate reductase (DHFR)-histidine triad nucleotide nanorings by chemically induced self-assembly. DHFR molecules with fused peptide chain of variable length were spontaneously self-assemble into protein macrocycles after treatment with a dimeric enzyme inhibitor, Bis-MTX-C9. The ring size, ranging in size from 10 to 70 nm, was dependent on the length and composition of the peptide linking the fusion proteins. The enzymatic efficiencies for the monomer and intramolecular macrocycle were found to be nearly identical, while the larger dimeric nanoring was found to have a modestly lower kcat/Km value. The nanorings catalytic efficiency was dependent on ring size, which indicated that the arrangement of supermolecular assemblies of enzymes may be used to control their catalytic parameters. However, the activator used for multi-enzyme assembly has not been reported before, which can greatly improve enzyme activity and may have greatly potential in mu lti-enzyme biosynthesis. Cofactor induced multi-enzyme assembly Cofactor-dependent enzymes, such as oxidoreductases and transferases, intramolecularly assembly of enzyme subunits by cofactor binding have been widely reported. Cofactor as a small molecular for enzyme catalysis. Cofactors can also be used for inducing multi-enzyme assembly. Bis-NAD+ has been reported for affinity precipitation of dehydrogenases in 1980s. Mansson et al used bis-NAD+ analogue to locate lactate dehydrogenase and alcohol dehydrogenase face to face and then cross-linked of the two enzymes with glutaraldehyde on agarose beads. The study of site-to-site directed immobilization effect improve the NADH production from 19% to 50%, which indicated that the NADH was preferentially channeled to lactate dehydrogenase due to the positioned active sites of the two enzymes. Similar work reported by Siegbahn as the bi-enzyme complex was formed by crosslinking lactate dehydrogenase and alcohol dehydrogenase with glutaraldehyde, which indicated an enhancement of 1.36 fold of the NADH regeneration when lactate dehydrogenase and alcohol dehydrogenase were site-to-site oriented. Cofactor induced assembly can form the site-to-site oriented structure, has the advantage easy operation and maintains the enzymes’ activity maintain. However, the interaction of NAD+ with enzyme is relatively low. Cofactor analogues have been reported for enzyme catalysis, which have the advance of low cost and high stability. The improvement of cofactor analogues for multi-enzyme assembly is promising. Metal ions induced multi-enzyme assembly Metal ions guide proteins into forming large assemblies, which provide a wide platform to modulate the metal coordination environment through distant, noncovalent interactions, exactly as natural metalloproteins and enzymes do. Metal ions in metalloenzymes located in the pocket whose shape fits the substrate, which are usually coordinated by nitrogen, oxygen or sulfur centers belonging to amino acid residues. Since approximately half of all proteins contain a metal ion, metal ions induced enzyme assembly is a promising method. Metal ions induced protein assembly is recently hot topic. There are two main types of metal ions induced protein assembly, namely, metal ions chelating sites on the artificial His-tags of enzymes and chelating sites on the surface of enzymes. His-tagging is the most widespread strategy to purify recombinant proteins. With the addition of 4-10 poly-histidine tag to the N terminus or C terminus of a target protein, the tagged protein purification was achieved by immobilized metal affinity chromatography. Multi-enzyme complex were formed with the Ni2+ and bis-His coordination of GDH-NOX fused enzymes, which enhanced enzyme activity and stability for the biosynthesis of DHA from glycerol with cofactor regeneration.. Chelating sites on the surface of enzymes The metal ions coordinated with the chelating sites on the surface of proteins was reported. Chelating sites should be on the surfaces to coordinate with metal ions, and the interfaces where chelating sites are located should be complementary to form stable self-assemblies. J Am Chem Soc 135, 10966-10969 (2013). ]reported a variant of glutathione S-transferase (sjGST-2His) which has two properly oriented His metal-chelating sites on the surface self-assembled in a fixed bending manner to form protein nanorings. The accurate orientation of proteins and self-assembly was based on metal-ion-chelating interactions and nonspecific protein–protein interactions. This work provides a de novo design strategy for the construction of novel protein superstructures. The self-assembly of glutathione S-transferase into nanowires was also reported[Zhang, W. et al. Self-assembly of glutathione S-transferase into nanowires. Nanoscale 4, 5847-5851 (2012).]. Designed metal coordination interactions to arrange enzyme into highly ordered supramolecular architectures has been reported recently[Salgado, E.N., Radford, R.J. Tezcan, F.A. Metal-Directed Protein Self-Assembly. Accounts Chem Res 43, 661-672 (2010).].Enzymes represent particularly attractive building blocks due to their chemical and structural versatility, for new and improved supramolecular properties. Metal-directed enzyme self-assembly yields stable architectures and high catalysis efficiency. These emergent physical and functional properties are attained with minimal modification of the original building blocks Brodin reported the self-assembly of a designed variant of cytochrome cb(562) by zinc ion coordination to uniform 1D nanotubes or 2D arrays with very high chemical stabilities. Biomacromolecules 12, 3400-3405 (2011).] the self-assembled inorganic–protein arrays by FeII complexation of protein-conjugated terpyridine units (ligand) to form well-defined and controllable size and structure. Residue-specific conjugation between the complexing unit (terpy) containing an activity-based probe and a corresponding active enzyme (papain) performed on this unique building block (ligand) leads to chemical species of unprecedented constitution. Metal ion induced assembly are controllable by environmental factors that affect the coordination or reactivity of the metal ion: the presence of the metal itself, external chelators, pH, and the solution redox state. Thus, metal ions can augment or provide all three essential properties of proteins as nature’s favorite build-ing blocks: structure, chemical reactivity, and stimuli- responsiveness. Metal ions are frequently found in natural protein-protein interfaces, where they stabilize quaternary or supramolecular protein structures, mediate transient protein-protein interactions, and serve as catalytic centers. Paralleling these natural roles, coordination chemistry of metal ions is being increasingly utilized in creative ways toward engineering and controlling the assembly of functional supramolecular peptide and protein architectures. Here we provide a brief overview of this emerging branch of metalloprotein/peptide engineering and highlight a few select examples from the recent literature that best capture the diversity and future potential of approaches that are being developed. Conclusions and Outlook Constructing functional multi-enzyme complexes in vitro by mimicking the natural enzyme complex has great biotechnological potentials in metabolic engineering, multi-enzyme-mediated biocatalysis, and cell-free synthetic pathway biotransformation. This review summarizes chemically assembling of multi-enzymes based on the affinity included by small molecular, namely, cofactor, substrate, inhibitor, and metal ions, et al. Distinctions were made based on the assembling driving force, structure of multi-enzyme complexes and mechanism of catalytic efficiency enhancement. Furthermore, the current challenges of multi-enzyme assembly in vitro induced by chemicals was addressed and gave an outlook on future developments. In this review, a classification of multi-enzyme assembly methods is proposed. Special emphasis is placed on the description of constructing functional multi-enzyme complexes by small molecular induced self-assembly. Assembling of multi-enzymes based on the affinity induced by small molecular, namely, cofactor, inhibitor, and metal ions were discussed. Furthermore, the advantage and disadvantage of each method from the reaction and process considerations are described. A variety of approaches for multi-enzymatic synthesis in vivo using biological systems or in vitro with isolated biocatalysts have been successfully used for the synthesis of complex molecules, especially the chiral chemicals which frequently are not readily accessible by chemical synthesis. In the long term, multi-enzyme processes will replace many chemically catalyzed processes. Biocatalysis today is growing not only in the fine chemicals and pharmaceuticals but also in the production of bulk chemicals. The relevant multi-enzyme catalysis processes have a significant potential for industrial application. Several challenges remain for multi-enzyme processes despite the strong drivers for greener and ever more effective chemical process technology.Multi-enzyme assembly into exquisite, complex, yet highly ordered architectures is challenging due to the complexity of enzyme structures and interactions. Consequently, the prediction of multi-enzyme complex configurations, the structure controlled assembly and the dynamic kinetic simulation of assembly process are also challenging. Current efforts aim at the prediction of multi-enzyme complex configurations as well as at nanoscale reconstruction, and control of cascade reaction. The design of multi-enzymatic systems based on the structure controlling and function prediction. In Nature’s hierarchy such design and engineering studies can provide useful information. New approaches that allow the controlled assembly of multiple enzymes at a nanometer scale with precisely structure and function will increase reaction rates and the efficie ncy of longer synthetic enzymatic cascades. Another frontier in multi-enzyme synthesis is the design of multi-step processes, involving mathematical modeling, process technology, and protein engineering. By viewing multi-enzyme assembly process in terms of structure and function relationship, it is possible to unify a diverse range of investigations, highlights their interrelationships, and see routes.

Thirty Years’ War The Thirty Years' War was a series of wars fought in the Holy Roman empire, involving most of the countries within, the war lasted 30 years. This war massively affected europe and created a cultural divide between catholics and protestants. Philip III of Spain attempted to continue the foreign policy views of his father, Philip II, which basically meant that Spain had to be kept ready for war. Thus in 1618, when Ferdinand II, heir apparent to the throne of Bohemia, began to slash certain religious privileges enjoyed by his subjects there, they immediately appealed for aid to the Protestants in the rest of the empire and to the leading foreign Protestant states: Great Britain, the Dutch Republic, and Denmark (Trueman). Even though the war between the German Catholics and German Protestants were all in the Holy Roman Empire were both Christian it slowly spread was more of a fight over the balance of power. There were two sides in the Thirty Years’ War the Protestant League and the Catholic League. The Protestant League consisted of Frederick of Bohemia who was the leader of the Protestant princes, Denmark led by king Christian IV, Sweden led by king Gustavus Adolphus, and the Netherlands led by the house of orange all financially supported by king of France Louis XIII. The Catholic League consisted of the leader Duke Maximillian of Bavaria, Holy Roman Empire Led by Emperor Ferdinand II, and king of Spain Philip IV (Billiet). There were five major phases of the Thirty Years’ War the Bohemian Revolt, Palatinate phase, Danish intervention, Swedish intervention, and the French intervention (Thomas). The German infantry failed horrendously, and the Spanish veterans held their ground due to some help from the musketeers in the woods. The spanish managed to wound Brunswick and drove his army off the field. The Protestants lost 3,000, 500 were wounded, and 1,500 captured. The spanish couldn’t have asked for a better fight. The Spanish suffered 300 dead and 900 wounded. The following day the Spanish cavalry obliterated the remaining Protestant infantry as they marched. Of the 24,000 men that left the Palatine that day only 3,000 Protestant cavalry managed to reach the next town (Thomas). As shown many times in historical past battles like the persians vs. the greeks numbers don’t matter its the strategy and will of the people to protect their way of life.

Reflection on Jack Hodgins ”By the River” Essay

In the story of By the River, the author relates the love story between the young woman, Crystal Styan, and her husband, Jim Styan, in the terms of faith, responsibility and dream. After reading this story, I wonder that why Jim chose to leave and why Crystal still believed he will come back, even though she realized that her dream of the life with him would not come true. When Crystal was in grade ten, she gave up the opportunity of studying further and chose to live a wonderful and idyllic life with Jim. After they moved to the log cabin which sits on the edge of river, they began to build a farm. Since Jim was lack of farming knowledge and had no any experience before, six chickens were frozen to death in the cold winter and the cow that he has just bought stepped off the train and ran away. He eventually abandoned his house and wife irresponsibly and left on the train after he cannot accept the fact that the dream of becoming famer will not come true. In contrast, Crystal always believed his husband and tried her best to help Jim realize his dream, even though sometimes she understood the decision that living with Jim was wrong. Crystal has always loved Jim so that she trusted her husband will be on the way home one day and came to the station to wait him for the past six month. But all are delusional and impossible, her husband will never return on the train, because the conductor told her that he was not on board. In conclusion, this kind of woman, Crystal, is frankly pathetic because her husband ruthlessly abandoned and left her without any words, while Jim is certainly looked down upon by others, because he is self-concern and irresponsible for their love and relationship.

Duck Dynasty´s Phil Robertson and His Interview and...

There is a fine line between expressing your personal opinion and verbally criticizing others. However, when it comes down to it, what was Phil Robertson, the â€Å"dad of Duck Dynasty,† really doing? â€Å"My mission today is to go forth and tell people about why I follow Christ and also what the bible teaches, and part of the teaching is that women and men are meant to be together,† he stated. Personally, I believe that Phil Robertson was being brutally honest and I see no issue with that. Arts Entertainment (AE) cannot punish him for his personal beliefs. Quite frankly, I think it was an unintelligent move on their part to ask a man of God, such as Phil, what his belief was on gay marriage. What else would you have expected him to say? â€Å"I myself am a product of the 60s; I centered my life around sex, drugs and rock and roll until I hit rock bottom and accepted Jesus as my Savior,† stated Robertson at one point in the interview. Regardless whether or not AE thought that his statements were derogatory, they certainly have many other hit TV shows with stars whose opinions differ from Phil’s. AE was quick to jump on him and suspend him indefinitely, which was extremely unfair for him. It seems to me that if he said what they had wanted him to say, he would be fake. Television is 85% fake these days and I cannot wrap my head around the fact that someone is being judged for being honest about one of their own personal beliefs.
